Brief Description of Research Interest :

Aspecific set of human genes is transcriptionally induced when cells areinfected with viruses or treated with interferons or dsRNA, the most prominentof which are the members of the ISG56 family. P56, the product of ISG56,interacts with human papilloma virus (HPV) E1 protein. E1 protein of HPV is oneof the key viral proteins that has enzymatic activities and is required forviral DNA replication. I have investigated the functional effect of P56interaction with the E1 protein. Transient DNA replication assays revealed thatHPV DNA replication is inhibited in a dose dependent manner by addition of IFN-b, which induces the expression of P56protein. Ablation of P56 expression by siRNA inhibited the effect of IFN.  Importantly, purified recombinant P56 proteincould inhibit HPV DNA replication invitro. I have identified the region of P56 that interacts with E1 proteinand inhibits viral DNA replication. Further studies with purified E1 proteinshow that P56 protein interferes with one of the key enzymatic activities of E1protein, the helicase activity. These findings demonstrate that interferonsensitivity of HPV replication is mediated by the IFN- inducible protein P56.
